Permutations count ordered arrangements: P(n,r) = n!/(n−r)!. Combinations count unordered selections: C(n,r) = P(n,r)/r!.

What's the difference between the two?

Permutations care about order (ABC ≠ BCA); combinations don't (ABC = BCA counts once).
